HOT - Botox as a Treatment for Depression

A new study to be published in the May issue of The Journal of Psychiatric Research finds that it may be possible to treat depression with Botox injections, which prevent patients from frowning.

Seventy-four patients with major depression were randomly assigned to receive either a Botox or placebo saline injection in a double-blind trial. (While there is evidence that the blind was at least partially breached, surprisingly only about half of the patients who received Botox injections guessed correctly. Furthermore, a patient's ability to discern which injection he or she received had no significant effect on treatment response.) Six weeks after the injection, 52 percent of the subjects who received Botox exhibited relief from depression, compared with only 15 percent of those who got the saline injection.

The idea that our facial expressions can enhance, perhaps even create, emotional responses isn’t new (Charles Darwin proposed the theory of “facial feedback hypothesis” way back in 1872: “The free expression by outward signs of an emotion intensi­fies it,” he wrote). Neither is the theory that, by inhibiting one's ability to frown, Botox can improve one's mood. Back in 2008, psychologists at the University of Cardiff in Wales provided an anxiety and depression questionnaire to 25 women, half of whom had received Botox injections. While those who received the Botox didn't say they felt any more attractive than they had before the injection, they did report feeling less anxious and happier (suggesting that Botox's ability to boost one's mood can't simply be attributed to its ability to boost one's appearance).

There are many reasons, the authors of the study to be published in The Journal of Psychiatric Research speculate, why Botox may help alleviate the symptoms of depression. "First, frowning may affect the way people feel about themselves when they look in the mirror and the way others respond to them," they write; Eliminating a person's ability to grimace will logically cause them to interact more positively with those around them, boosting their mood.

Finally, and most dramatically, the authors hypothesize that the act of frowning in itself may be a depressant. In other words, our brain monitors "the extent of our facial muscle contraction and muscle tension" (i.e.our facial expressions) and adjusts our mood accordingly; Muscles tensed up into a grimace will cause the brain to generate the corresponding emotional experience. Botox treatment, the authors predict, "would interrupt the normal circuitry, reduce distress signals to the brain and thereby influence mood in a favorable way."

Botox or no Botox, the study illustrates that our facial expressions are important. Choosing to give into our dismal mood and frown – instead of smiling through any internal unpleasantness – may impact our ability to adopt a sunnier outlook.

Not only that, but our emotions are highly contagious. Because we tend to mimic the behavior of those around us, we are highly susceptible to "catching" each other's' moods. Both negative and positive emotion can spread through an office much like a cold does.

Maybe the solution is Botox for everyone. Or perhaps it's simply to heed the advice we all got (and rolled our eyes at) as children. The next time you face a rainy Monday commute, go ahead and try it: Turn that frown upside down and see what happens.

Antioxidant Tiron Is An Anti-Aging Skin Miracle

Tiron- the antioxidant can protect the skin against sun damage and even prevent premature aging.

The study was conducted by researchers at Newcastle University. The team said that Tiron can provide "100% protection against mitochondrial DNA damage."

Antioxidants are natural or man-made compounds that delay cell damage. Fruits and vegetables contain several types of antioxidants such as beta carotene, lutein and lycopene.

As our skin is exposed to more and more of the sun’s rays, especially UVA and UVB radiation, DNA within skin cells are broken down due to oxidative stress, which causes skin’s elasticity to wear. A study out of Newcastle University revealed the antioxidant Tiron could be the anti-aging miracle we need to keep our skin looking younger, even into old age.

Ultraviolet A radiation has the ability to penetrate deep into the skin and damage the DNA by generating free radicals. The best antioxidants prevent this destruction by targeting the mitochondria or the "power house of the cell."

For the study, researchers looked at the efficacy of known anti-oxidants such as resveratrol (found in red wine) and curcumin (found in curry) and compared it with the latest compound, according to news release.

The team first treated skin cells with antioxidants and exposed them to UV A radiation at doses that humans normally experience on a summer's day.

They then used polymerase chain reaction to copy the skin cell DNA and look at the levels of change in the genetic material.

The team found that Tiron- 4, 5-Dihydroxy-1, 3-benzenedisulfonic acid disodium salt monohydrate- provided a 100 percent protection against mitochondrial DNA damage. However, the wine compound resveratrol and turmeric compound curcumin provided just 22 percent protection.

But, you won't be seeing Tiron in food or cosmetics soon because it still needs to pass several tests including a toxicity test.

"To discover that Tiron offers complete protection against UVA damage is exciting and promising, however, it is early days as Tiron is not a naturally occurring compound and has not yet been tested for toxicity in humans although there have been a few studies on rats," said Mark Birch-Machin, Professor of Molecular Dermatology at Newcastle University.

The study was funded by Biotechnology and Biological Sciences Research Council (BBSRC) and Unilever and is published in The FASEB Journal.


Although ultraviolet B radiation (UVB) is considered the leading cause of skin reddening and sunburn, recent studies show that ultraviolet A radiation (UVA) has the ability to penetrate the skin's surface, damaging our skin through oxidative stress. Oxidative stress, also known as free radical stress, is caused by an imbalance between reactive oxygen species that results in the deterioration of DNA and ultimately collagen, elastin, and the skin’s supportive fibers. When collagen in the body is broken down, the skin will start to lose its elastic quality, causing wrinkles.

"To discover that Tiron offers complete protection against UVA damage is exciting and promising, however, it is early days as Tiron is not a naturally occurring compound and has not yet been tested for toxicity in humans although there have been a few studies on rats," said Mark Birch-Machin, professor of molecular dermatology at Newcastle University, in a press release.

Birch-Machin and his colleagues from the university compared Tiron’s effect on UVA radiation exposure and free radical stress against other antioxidants, including resveratrol found in red wine, turmeric found in curry, lycopene found in tomatoes, antioxidants found in green tea, and cosmetic creams. The research team focused on each antioxidant’s effect on mitochondria, also known as the batteries of skin cells.

Researchers treated skin cells with each of the antioxidants and exposed them to a dose of UVA equivalent to what we would experience on a warm summer’s day. Skin cell damage was assessed by copying DNA through a polymerase chain reaction machine. Tiron was able to provide 100 percent protection against mitochondrial DNA damage, including 100 percent UVA protection and 100 percent protection against oxidative stress.

Other antioxidants that were compared against Tiron did not have the same success rate with preveting UVA damage. Resveratrol was only able to protect the skin against 22 percent of both UVA and oxidative stress, and curcumin offered eight percent protection against UVA and 16 percent oxidative stress prevention. Researchers say the next step in developing a viable Tiron based sunscreen will be testing the toxicity of compounds based off of the antioxidant’s makeup.

"This finding on Tiron provides us with a platform to study an antioxidant - preferably a naturally occurring compound with a similar structure which could then be safely added to food or cosmetics," said Dr. Anne Oyewole, also from Newcastle University.

According to the Skin Cancer Foundation, approximately 95 percent of UV radiation that reaches Earth’s surface is UVA. UVB radiation may be more intense than UVA, but we are exposed to UVA 30 to 50 times more than UVB. Tanning beds, which emit UVA doses as much as 12 times that of the sun, have been linked to a 75 percent increase in melanoma risk among teenagers and adolescents. Melanoma, the deadliest form of skin cancer, causes over 8,000 deaths in the United States each year.
